Objectives:To determine if a phosphodiesterase type 5 (PDE5) inhibitor is effective in reducing the adverse effects of the nitrate-dependent vasodilator vasodilator papaverine (PDE5). PDE5 inhibitors are a class of drugs with the active ingredient nitrite (NO) in a nonselective manner.
Methods:Twenty-four-day, placebo-controlled, crossover single-blind, randomized, single-center study in which a PDE5 inhibitor, papaverine (1, 5-mg) was administered to healthy male volunteers. In the first study, a single oral dose of papaverine (0.25, 1.25, and 1.5 mg) was administered at 0, 2, and 4 h.
Results:Papaverine decreased plasma levels of papaverine by 90.7% in comparison with baseline (p < 0.001), which was significantly less in subjects receiving the highest dose of papaverine (1.25 mg) than the lowest dose (0.25 mg) (p < 0.001). papaverine (1.25 mg) significantly decreased the plasma levels of papaverine by 94.9% (p < 0.001) in the placebo group and 94.5% (p < 0.001) in the papaverine group.
Conclusions:The effects of papaverine on papaverine plasma levels are similar to those seen with papaverine-containing medications.
